Lions mane agar tek?
« on: December 20, 2013, 06:48:40 AM »
Is there a special way to put lions mane on agar?
I am getting bacterial contaminations on all of my plates. It occurs around the piece of mushroom I put in the agar.
I have read that lions mane is known to harbour bacteria and you need to pour hot age over the culture after you have put it on agar. I tried this but the piece of mushroom let's go of the agar and then floats off center.

Has anyone got any clear instructions or a better method?

Would peroxide agar help?

Hmmm maybe not because that's mostly used to prevent spores from germinating.

Could you let the mycelium grow and then transfer away a piece from the leading edge?  A piece that appears to not have contam on it?  It's not a guarantee, just an idea.

Could I dip the mushroom in peroxide then put it on the plate.
I have 50% peroxide it's pretty strong stuff

I think that could be a good idea.  It's definitely worth the experiment.

Is the contam all over the plate?
Are there any sections of clean myc that are outrunning the contam?
Could you take a clean wedge and re-plate it to clean it up?

Very interested to hear how you go as I'd love to grow lions mane in the very near future.
